中新社合肥7月10日电 (吴兰 张淑凡)亿万条直径仅为头发丝几十分之一的神经纤维如何快速看清、看全?中国科学技术大学毕国强教授、刘北明教授与合作者创新性开发出一套“超级CT扫描仪”,给出解决方案。
据悉,这个三维成像技术可清晰捕捉直径为数微米的单根神经纤维。相关成果于北京时间7月10日发表于国际学术期刊《细胞》(Cell)。
在生物体内,一张由亿万周围神经纤维编织成的精密“生命物联网”无处不在,承载着大脑与全身器官的双向通讯,调控运动、感觉以及内脏功能。然而,看清这张网络的全貌却极其困难——神经纤维直径仅为发丝几十分之一,蜿蜒交织遍布全身。
长期以来,受限于成像速度与分辨率矛盾,“如何同时‘看得清’单根神经纤维,又能‘看得全’全身神经网络的整体构架”,是神经科学领域悬而未决的技术难题。
研究团队创新性开发出一套超高速小鼠全身亚细胞分辨率三维成像技术“blockface-VISoR”,如同为生命体构建一套“超级CT扫描仪”。该技术仅用40小时即完成成年小鼠全身成像,获取单通道约70 TB原始图像数据(相当于数千部高清电影),效率较现有技术提升数倍至数十倍,可清晰捕捉直径数微米的单根神经纤维。
“blockface-VISoR”技术采取“样品原位切片+切面三维成像”策略,整合了精密振动切片装置与“同步飞扫”三维显微成像系统,对透明化包埋的小鼠样本分层扫描,每次拍摄约0.6毫米深度的三维图像后,自动切削0.4毫米组织,再通过智能算法将数百层图像无缝拼接。
“眼见为实,我们可以不依赖间接证据,直接‘看见’全身神经网络如何精密连接、如何与不同组织器官相互作用。”通讯作者毕国强说。
据介绍,这一突破性研发的全球最快的小动物全身亚细胞分辨率三维成像技术,首次实现了小鼠全身神经网络精细三维图谱高清绘制。此外,团队将持续公开研究成果图像数据集,实现资源全球共享,以推动生物医药交叉合作。(完)
