中新网杭州9月5日电(张煜欢)近日,浙江大学医学院附属邵逸夫医院心内科张文斌教授团队与特聘副研究员王雅楠在国际权威期刊《Bioactive Materials》上发表一项研究成果,该研究成功开发出一种微环境响应型血管支架涂层,能够智能调控支架植入后的凝血—炎症相互作用,显著促进血管修复与再生。该成果也已获得国家发明专利授权。
冠状动脉粥样硬化性心脏病是全球范围内致残和致死的主要病因之一,而血管支架作为血运重建的关键器械,虽能迅速恢复血流,却常常面临术后再狭窄、血栓形成、持续性炎症等问题。
随着社会老龄化加剧,心血管疾病患者数量不断攀升,开发兼具良好生物相容性和功能响应性的新一代支架,成为临床治疗的迫切需求。

张文斌研究团队设计出一款可响应病变微环境的智能支架涂层。该涂层具备双重调控机制。首先,在炎症环境中,涂层通过感应高表达基质金属蛋白酶9,精准释放丹酚酸A,有效清除自由基、调节免疫细胞行为,并抑制平滑肌细胞过度增殖;其次,可同步持续释放抗凝药物替罗非班,阻断血小板聚集和血栓形成,从根本上干扰“凝血—炎症”恶性循环。
该设计不仅在时间维度上覆盖了支架植入后的全程组织反应阶段,还在功能上同时兼顾抗凝、抗炎和促血管修复三大目标。
在动脉粥样硬化兔模型中,该涂层支架表现出优异的治疗性能:包括显著抑制内膜增生,改善血管结构与功能;减少血小板—纤维蛋白复合物及血小板—单核细胞聚集体的形成;降低脂质沉积,促进健康内皮层的再生与修复。
该研究不仅提供了一种全新的涂层技术策略,也为血管支架的下一代升级指明了方向。团队表示,未来将进一步推进该技术向临床转化,并结合材料学、生物信息学及人工智能方法,开发可实时监测血管状态、自动调节药物释放的“智能支架系统”,实现对不同患者、不同病变阶段的个体化精准治疗。
随着这项技术的成熟与推广,有望显著提升冠心病患者介入治疗的中远期疗效,改善生存质量,同时降低再干预率,为健康老龄化战略提供技术支撑。(完)
